Real time RT-PCR, Western Blotting ,Apoptosis detection and Chromatin immunoprecipitationDown-regulation-RUNX3 is a gastric and colon tumor suppressor, functioning downstream of TGF-beta.

RUNX3 downregulates c-Myc expression by two parallel pathways-directly at the transcriptional level and through attenuation of beta-catenin/TCFs, downstream of BMPs in colorectal cancer cells.

Upregulation of RUNX3 by BMP reduces c-Myc expression.

TaqMan assayMethylation-TaqMan assay used to genotype rs4939827 and logistic regression to assess the association of rs4939827 with risk of CRC.

Minor allele (G) of the germline intronic SMAD7 variant rs4939827 is associated with a lower risk of CRC with earlier tumor stage and CRC without methylation of the tumor suppressor RUNX3.

Gene Type protein coding
Gene Family -
Description This gene encodes a member of the runt domain-containing family of transcription factors. A heterodimer of this protein and a beta subunit forms a complex that binds to the core DNA sequence 5'-PYGPYGGT-3' found in a number of enhancers and promoters, and can either activate or suppress transcription. It also interacts with other transcription factors. It functions as a tumor suppressor, and the gene is frequently deleted or transcriptionally silenced in cancer. Multiple transcript variants encoding different isoforms have been found for this gene. [provided by RefSeq, Jul 2008]
